Synthesis of DSPE-PEG-Angiopep-2
To synthesize DEPE-PEG-Angiopep-2, 5 mg of DSPE-PEG-Mal was completely dissolved in 10 mL of water. Then, 6 mg of Angiopep-2-cys was added to the solution, and the mixture was stirred at 37°C for 12 hours. The solution was dialyzed (MW3500) to remove excess Angiopep-2-cys. The resulting purified product, DSPE-PEG-Angiopep-2, was then freeze-dried and stored at ?20°C for subsequent usage.
DSPE-PEG-Angiopep-2的合成

CG/SN-38納米粒子(NP)的制備與之前報(bào)道的相同,但經(jīng)過了修改。G和1 mg SN-38溶解在20μL DMSO中,然后向溶液中加入1 mL水,然后超聲處理5分鐘。將制劑用蒸餾水(1L)透析(MW3500)12小時(shí)。然后,將NP溶液與一定量的巨噬細(xì)胞膜超聲2分鐘,以獲得巨噬細(xì)胞膜/IGC/SN-38(M-NP)。此外,通過超聲處理(400 W,2 s開/3 s關(guān),2 min)M-NP和預(yù)先制備的100μg DSPE-PEG-Angiopep-2,獲得1 mL Angiopep-2修飾的M-NP(AM-NP)。
