cy染料活化基團(tuán)的選擇

根據(jù)需要標(biāo)記的抗原、抗體、酶、多肽等分子所帶的可標(biāo)記基團(tuán)的種類（氨基、醛基或巰基）以及分子的酸堿性，選擇相應(yīng)的活化染料和反應(yīng)條件。為減少空間位阻影響，可在cy染料與被標(biāo)記物之間加入交聯(lián)臂結(jié)構(gòu)。常見抗體、抗原、酶、多肽對(duì)應(yīng)的染料活化基團(tuán)如下：

1.標(biāo)記氨基的活化基團(tuán)：NHS ester (N-羥基丁二酰亞胺酯)。

2.標(biāo)記醛基的活化基團(tuán)：Hydrazide酰肼。多的用于神經(jīng)元細(xì)胞的電極失蹤和神經(jīng)縫隙連接研究

3.標(biāo)記巰基的活化基團(tuán)：Maleimide，馬來酰亞胺 

近紅外染料系列產(chǎn)品（CY系列/IR系列）：

近紅外染料和傳統(tǒng)的可見光染料相比有較大的優(yōu)勢(shì)。由于細(xì)胞和組織的自發(fā)熒光在近紅外波段小，因此在檢測(cè)復(fù)雜生物系統(tǒng)時(shí)，近紅外染料能提供更高的特異性和靈敏度。同時(shí)，由于光波在近紅外區(qū)段的組織透過性更好，因此近紅外染料尤其適合體內(nèi)熒光成像（活體成像）這一近幾年迅速發(fā)展的新興領(lǐng)域。另外，近紅外染料也是細(xì)胞內(nèi)/全細(xì)胞免疫印記和傳統(tǒng)基于膜介質(zhì)免疫印記的理想染料。
